Their chemical structures were determined based on extensive spectroscopic studies and chemical methods. The aglycones of pavitnoside B and pavitnoside C were not reported in previous work. The cytotoxicity of all saponins was evaluated against human glioblastoma U87MG and U251 cell lines. The new spirostanol saponin 1 displayed weak anti-proliferative activity against U87MG cell line and the known saponins 8 and 9 exhibited significant cytotoxicity against the two tumor cell lines, with IC50 values of 2.16 to 3.14 μM, but did not affect the growth of primary cultures of human astrocytes. 摘要: 從越南梧桐根莖中分離到4個(gè)新的螺甾醇類皂苷,命名為pavitnosides A-D(1-4), 6個(gè)已知甾體類皂苷5-10。它們的化學(xué)結(jié)構(gòu)是根據(jù)廣泛的光譜研究和化學(xué)方法確定的。前文未報(bào)道過(guò)其苷元B和C的苷元。評(píng)價(jià)了各皂苷對(duì)人膠質(zhì)母細(xì)胞瘤U87MG和U251細(xì)胞系的細(xì)胞毒性。新合成的螺甾醇皂苷1對(duì)U87MG細(xì)胞系具有較弱的抗增殖活性,而已知的皂苷8和皂苷9對(duì)兩種腫瘤細(xì)胞系具有明顯的細(xì)胞毒性,IC50值為2.16 ~ 3.14 μM,但對(duì)人星形膠質(zhì)細(xì)胞原代培養(yǎng)物的生長(zhǎng)沒(méi)有影響。 該論文中,人膠質(zhì)瘤細(xì)胞系U251和U87MG的體外培養(yǎng)是使用Ausbian特級(jí)胎牛血清完成的。欲了解或購(gòu)買Ausbian特級(jí)胎牛血清可以聯(lián)系北京締一生物400-166-8600. |
